Taking omeprazole and sertraline together can increase the amount of sertraline in your bloodstream. This may increase the risk of side effects from sertraline, such as feeling sick (nausea), tremors, sleep problems, or feeling agitated.
In rare cases, it could lead to a serious condition called serotonin syndrome, which causes high body temperature, sweating, and confusion.

Why this happens
Sertraline is broken down in the liver by certain proteins (enzymes). Omeprazole can slow down these proteins, which means the sertraline stays in your body longer and reaches higher levels than intended.
